چکیده

Alkali-extracted erythrocyte ghost membranes from Rho(D)-positive and Rho(D)-negative donors were incubated with human immune anti-Rho(D) IgG and nonimmune IgG. After sensitization with IgG, the integral membrane proteins were solubilized in Brij 36T nonionic detergent and chromatographed by gel filtration. There was a distinct resolution of IgG into free and membrane-complexed forms. The IgG-complexed membrane proteins were isolated by the use of a staphylococcal protein A affinity support. The protein A-bound complexes were examined for polypeptide composition by gel electrophoresis after elution. Only Rho(D)-positive membrane proteins incubated with immune anti-Rho(D) IgG revealed intact band 3. Control Rh-negative membrane proteins that had reacted with immune anti-Rho(D) IgG and the Rh-positive membranes that had reacted with nonimmune IgG showed only low molecular weight fragments of band 3 that bound nonspecifically to IgG. Arguments are presented supporting a band 3 localization for the Rh antigen.

Antigenic determinants of senescent antigen of human erythrocytes are located in sialylated carbohydrate chains of Band 3 glycoprotein.

Naturally occurring IgG autoantibody against Band 3 glycoprotein of human erythrocyte membrane (anti-Band 3) recognizes the erythrocytes modified with oxidizing or SH-blocking agents as well as senescent erythrocytes. Location of the antigenic determinants of Band 3 this autoantibody recognizes was investigated by competitive inhibition studies of the antibody binding to the modified cells. Aut...

Ultrastructural mapping of methyldopa and anti-D IgG erythrocyte antigen receptors.

The ultrastructural distribution pattern and site density of alpha-methyldopa immunoglobin G (alpha-MD IgG) on the red cell membrane was observed and compared with that of anti-D IgG, with ferritin-conjugated rabbit anti-human IgG and [125I]anti-D. alpha-MD IgG binds to all common types of human red cells, both Rho (D) positive and negative, to give a random, aperiodic distribution pattern gros...

Band 3 protein: structure, flexibility and function.

The electroneutral exchange of chloride and bicarbonate across the human erythrocyte membrane is facilitated by Band 3, a 911 amino acid glycoprotein. The 43 kDa amino-terminal cytosolic domain binds the cytoskeleton, haemoglobin and glycolytic enzymes. The 52 kDa carboxyl-terminal membrane domain mediates anion transport. The protein is a functional dimer, in which the two subunits probably in...

Naturally occurring human anti-band 3 autoantibodies accelerate clearance of erythrocytes in guinea pigs.

A variety of naturally occurring autoantibodies (NOAs) have been found in sera of animals and humans. Although their specific homeostatic role in the clearance of altered or senescent cells has been proposed and in vitro studies support such functions, in vivo evidence has been lacking. We studied the effect of affinity-purified human anti-band 3 NOA on the survival of untreated and diamide-tre...

A carbohydrate-deficient membrane glycoprotein in human erythrocytes of phenotype S-s-.

1. We investigated the membranes of human erythrocytes which completely lack the blood-group antigens S and s (denoted as S-s-) as part of a study of the structure and function of the surface glycoproteins of the human erythrocyte. 2. The S-s-erythrocyte-membrane glycoprotein PAS-3 band was much less intensely stained in comparison with that of the glycoprotein from normal erythrocyte membranes...
